Why Should I Take Tetrahydromagnolol?

Tetrahydromagnolol is a cutting edge derivative of one of the main bioactives in magnolia bark, magnolol. Tetrahydromagnolol is a major metabolite that gets formed after magnolol is orally supplemented, and presents with a very interesting effects profile! However, the metabolism of magnolol is unpredictable, and thus we have taken the guesswork out of it, and hydrogenated magnolol in order to create pure tetrahydromagnolol. This compound now can get to work fast, and exert potent effects!

Tetrahydromagnolol is fascinating because it has significant agonist effects at the cannabinoid CB2 receptor and mild agonist effects at the cannabinoid CB1 receptor. Additionally, it also acts as a significant antagonist at the novel GPR55 cannabinoid receptor. These receptor targets produce a very interesting effects profile that is rarely seen, and offer unique effects on pain management, inflammation, gastrointestinal health, and mood!

We should also mention that tetrahydromagnolol is incredibly rare! It took us years to develop this product, and as of 2023, we are the only vendor that carries this unique compound!

What is Tetrahydromagnolol?

Tetrahydromagnolol is one of the primary metabolites of magnolol, which is a highly bioactive lignan found in the bark of Magnolia officinalis. When magnolol is taken orally, it becomes metabolized by various enzyme systems, which then yields tetrahydromagnolol as one of the primary metabolites. Tetrahydromagnolol is even more bioactive, and most interestingly becomes a much more potent and selective cannabinoid CB2 agonist!

That being said, tetrahydromagnolol formation in humans after oral administration of magnolol, is not going to be very efficient. First of all, a large portion of magnolol is excreted unchanged, which drastically reduces the potential for tetrahydromagnolol formation. Furthermore, tetrahydromagnolol is only one of a few possible magnolol metabolites, and the metabolism is being facilitated by highly variable enzyme systems. Due to this, the amount of tetrahydromagnolol formed after oral ingestion of magnolol is likely going to be fairly low. This is why “pre-metabolized” tetrahydromagnolol is very interesting, as you can immediately and reliably access the wonderful benefits of it!

Tetrahydromagnolol is made by a fairly lengthy process, starting with the harvesting of Magnolia officinalis bark. From the bark, we can extract and isolate magnolol. Once the magnolol has been highly purified, it is reacted with molecular hydrogen, in a process known as hydrogenation. This then removes the double bonds in magnolol, and yields tetrahydromagnolol!

Next Generation Magnolia Bark

Magnolia bark is well known for its ability to induce relaxation, boost mood and modulate pain. These effects can be attributed to two different compounds, magnolol which is more active on the endocannabinoid side, and honokiol which is more active on the GABAergic side. When either of these compounds are hydrogenated, their respective affinities drastically increase. For tetrahydromagnolol, this means that it is about 19 times more active at the CB2 receptor than its parent compound, magnolol. So what does this mean in terms of effects?

The CB2 cannabinoid receptor is a very unique receptor which is widespread throughout our body and central nervous system. Within our body, it interacts a lot with immune cells, especially those which control inflammation. In fact, there is a particularly high concentration of CB2 receptors in the gastrointestinal tract, and due to this, CB2 agonists have the ability to greatly dampen inflammation throughout the gastrointestinal tract. Natural CB2 agonists are fairly hard to come by though, especially those which possess high potency like tetrahydromagnolol. Interestingly enough, within traditional Chinese practices, Magnolia officinalis bark has been used for centuries for alleviating gastrointestinal distress! Thus, based on the pharmacological enhancement of magnolol activity via hydrogenation, tetrahydromagnolol should be a fantastic ally for those looking to enhance gastrointestinal comfort, via its potent CB2 agonist effects!

Another use for Magnolia officinalis bark in both traditional Chinese and Japanese practices, has been to use it for pain. This is interesting, because CB2 receptors have a potent regulating effect on pain signaling. With this in mind, the significant increase in CB2 affinity with tetrahydromagnolol, will make it a fantastic pain management supplement. Anecdotally, we have noticed robust pain management effects with tetrahydromagnolol which kick in quite rapidly. Research wise, there is a huge search for selective CB2 agonists, as they are non-psychoactive and can provide very significant pain regulating effects. Within this currently large body of research, there is also a lot of emphasis on CB2 agonists being particularly useful for nerve related pain.

On the topic of pain and research, there is currently also a lot of research ongoing on the GPR55 receptor. This is a novel cannabinoid receptor that was only discovered about two decades ago and there is still relatively little known about it. However, in the last few years, some relatively selective GPR55 ligands have demonstrated that antagonism of this receptor can also produce robust inflammation regulating effects. As luck would have it, tetrahydromagnolol is a relatively potent antagonist at the GPR55 receptor, which further adds to its pain management effects profile!

Another traditional use for Magnolia officinalis bark, has been for mood. Most of this activity likely is coming from the more GABAergic side of magnolia bark, however, the CB2 receptors can also control various aspects of mood. The bulk of these effects come from the fact that agonism of CB2 receptors in the brain produces a fantastic inflammation dampening effect. High levels of inflammation have been linked to low mood and can even have negative impacts on general cognitive function. Thus mitigating this with tetrahydromagnolol can produce a nice mood and cognition boosting effect!
